Protein structures in the GPR6 film (homepage, theme 01)
Rendered by us from atomic coordinates in the RCSB Protein Data Bank. GPR6 with the G protein Gs: PDB 8TYW (cryo-EM); GPR6 with CVN424: PDB 8T1W (crystal structure; the fusion protein used for crystallisation removed); both Barekatain M, Johansson LC, Lam JH, et al., Cherezov V. Structural insights into the high basal activity and inverse agonism of the orphan receptor GPR6 implicated in Parkinson's disease. Science Signaling 2024;17:eado8741. The receptor changes between these two measured structures. The Gαs helical domain, not modelled in 8TYW, is taken from PDB 3SN6 (Rasmussen SGF et al. Nature 2011;477:549–555), which also gave the membrane orientation. The Gαs subunit that travels from GPR6 to the cyclase changes between its receptor-bound structure (8TYW, with 3SN6) and its GTPγS-bound structure on the cyclase: adenylyl cyclase 9 with activated Gαs, PDB 6R3Q, with the active site located from 6R4O (Qi C, Sorrentino S, Medalia O, Korkhov VM. Science 2019;364:389–394); striatal neurons mainly use adenylyl cyclase 5, for which this structure stands in. Membrane orientations from the OPM database (Lomize MA et al. Nucleic Acids Research 2012;40:D370–D376). Small molecules (CVN424, cAMP) from the PDB Chemical Component Dictionary. The lipid bilayer is built procedurally; the timing of the relay, rates and amounts are illustrative.
